Research subject: Dissection of molecular mechanisms of somatic cell reprogramming



Recently the technology to generate pluripotent stem cells from somatic cells has been developed. These induced pluripotent stem cells (iPSCs) have the potential to be used extensively in medicinal areas, such as regenerative medicine, toxicological testing and drug screening. However, the majority of current reprogramming techniques involve viral gene delivery that results in multiple genome modifications. Although reprogramming strategies which do not involve genetic modification have been developed, the reprogramming efficiency of these strategies is extremely low, and not sufficient for routine use.

To improve the reprogramming technology and assure the safety of this technology for medical use, it is essential to understand the molecular mechanisms of the reprogramming process. Therefore, our laboratory aims to address the following questions:

> How does the gene expression profile change during the process of reprogramming.
> What are the key gene expression changes required for successful reprogramming?
> What are the major hurdles to successful reprogramming?
> How does chromatin modification and structure change during the successful reprogramming?

We are seeking enthusiastic postdocs who will enjoy working with us in the exciting field of reprogramming. Successful candidates should preferably have at least one first author paper in a major research journal and experience in all or some of the followings:

> Chromatin immunoprecipitation and interphase FISH
> Recombineering and gene targeting
> Live cell imaging, single cell tracking

Positions can commence from December 2010, and are for 2 years with the possibility of extension of contract for an additional 3 years.
